This monoclonal antibody is specific to estrogen receptor alpha (ER alpha) and shows minimal cross-reaction with other members of the family. ER is an important regulator of growth and differentiation in the mammary gland. Presence of ER in breast tumors indicates an increased likelihood of response to anti-estrogen (e. g. tamoxifen) therapy. It strongly stains nuclei of epithelial cells in breast carcinomas. Elastin is a polymeric protein and is the main component of the extracellular matrix of arteries. It is synthesized and secreted as a soluble, single-chain protein (tropoelastin), which undergoes a number of post-translational modifications prior to its organization into an elastic fiber in the extracellular space. Elastin performs a regulatory function during arterial development by controlling proliferation of smooth muscle and stabilizing arterial structure. It imparts elasticity to the connective tissue. During aging, the elasticity of connective tissue is reduced because of the cross-linking of collagenous fibers with elastin. This MAb recognizes a protein of 56 kDa, identified as cytokeratin 6 (CK6). In humans, multiple isoforms of Cytokeratin 6 (6A-6F), encoded by several highly homologous genes, have distinct tissue expression patterns, and Cytokeratin 6A is the dominant form in epithelial tissue. The gene encoding human Cytokeratin 6A maps to chromosome 12q13, and mutations in this gene are linked to several inheritable hair and skin pathologies. Keratins 6 and 16 are expressed in keratinocytes, which are undergoing rapid turnover in the suprabasal region (also known as hyper-proliferation-related keratins). Keratin 6 is found in hair follicles, suprabasal cells of a variety of internal stratified epithelia, in epidermis, in both normal and hyper-proliferative situations. This antibody recognizes a protein of 145 kDa, identified as CD11c. CD11c (ITGAX), a member of the leukointegrin family, shares the same beta subunit with other members of the leukocyte adhesion molecule family, which includes CD11a (LFA-1), CD11b (MAC-1) and CD11d (ITGAD), but has a unique alpha chain. CD11c has been shown to play a role in phagocytosis, cell migration, and cytokine production by monocytes/macrophages as well as induction of T-cell proliferation by Langerhans cells. CD11c is expressed prominently on the plasma membranes of monocytes, tissue macrophages, NK cells, and most dendritic cells (DCs). A lower level of expression is also observed on neutrophils as a result of its high level of expression on most DCs. An antibody to CD11c may aid in identification of lesions with histiocytic origin. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. This antibody recognizes a protein of ~140 kDa, identified as N-Cadherin (NCAD), also known as CD325. NCAD is a member of the Cadherin superfamily, and consists of five extracellular repeats, a transmembrane domain and a cytoplasmic domain. CD325 deficient mice die at day 10 of gestation and embryos display major heart defects and malformed neural tubes and somites. Consistent with this, CD325 has been implicated in several aspects of cardiac development including the precardiac mesoderm, establishment of left-right symmetry and cardiac looping morphogenesis. Furthermore, CD325 is normally involved in inducing cell cycle arrest and its expression is frequently deregulated in cancer cells. This MAb recognizes human 17-26 kDa protein, which is identified as cytokine TNF-alpha (Tumor Necrosis Factor-alpha). TNF-alpha can be expressed as a 17 kDa free molecule, or as a 26 kDa membrane protein. TNF-alpha is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha causes cytolysis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production. Recognizes a disaccharide epitope, Gal 1-3GalNAc, of Thomsen-Friedenreich (TF) antigen. S100A and S100B proteins are two members of the S100 family of calcium binding proteins. S100A is composed of an alpha and a beta chain whereas S100B is composed of two beta chains. This antibody is specific against an epitope located on the alpha-chain (i.e. in S-100A and S-100B) but not on the beta-chain of S-100 (i.e. in S-100B). This antibody can be used to localize S-100A in various tissue sections. Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker.
